She hailed from a well-to-do family; her father, Prabhakar Shilotri, was a prominent banker who founded the Shilotri Bank in Bombay, and her mother, Rattan Bai, was herself an actress in Marathi cinema. She studied at Cathedral School and later Baldwin Girls High School in Bangalore, following her family's relocation due to financial setbacks.
Shobhna Samarth made her film debut in 1935 with "Nigahen Nafrat," a bilingual Urdu-Marathi film, which though not commercially successful, earned her critical acclaim. She acted in a number of films through the 1930s and 1940s, working with various studios and starring in important films such as "Kokila" (1937), "Do Diwane" (1936), and "Pati Patni" (1939).

Her most defining role came as Sita in the mythological films "Bharat Milap" (1942) and "Ram Rajya" (1943), where her portrayal alongside Prem Adib as Ram made them iconic figures associated with these characters. Shobhna Samarth married Kumarsen Samarth, a filmmaker who had studied cinematography in Germany. She continued her career post-marriage, which was uncommon for women in her social milieu then.
Later, she transitioned into producing and directing films, notably launching the careers of her daughters Nutan and Tanuja, both of whom became celebrated actresses in Indian cinema. Some of her directorial works include "Hamari Beti" (1950) and "Chhabili" (1960).
